Roy was known for performing while standing motionless & wearing black clothes to match his dyed black hair & dark sunglasses. Roy had lots of hits in the 60's such as "Oh, Pretty Woman", "Crying", "Only The Lonely", "Running Scared", "In Dreams", "Love Hurts", "Leah" etc. In 1988, he co-founded the rock supergroup "Traveling Wilburys" with George Harrison, Bob Dylan, Tom Petty, & Jeff Lynne. They had a lot of great songs such as "Handle With Care", "End Of The Line" etc.
Though the definition is rather wide in the scope of what constitutes a bayou, in this case he is most likely referring to Louisiana. The bayou is a large, swampy lowlands where wildlife, especially fish, inhabit. You will also find alligators, snakes and many, many birds and insects. Small crustaceans called Crayfish are harvested from there. The water, while not stagnant, is very slow moving so you end up with moss, vines and lush, water loving plants and trees.
A bayou is a marsh or swampy body of water found along the southern Gulf Coastal states of Arkansas, Louisiana or Texas. While I do love Roy’s laidback version of this song, and we have him to thank for writing it, Linda Ronstadt’s recording is the far more well known (and in my humble opinion - better) performance. Her crystal clear, powerful, belting vocals will leave you stunned!
The operatic twin songs "Running Scared" and "It's Over" must have a go. Perhaps check "In Dreams" the original with a clip from David Lynch's "Blue Velvet" where the song appears subversively, which was instrumental in helping Roy Orbison's comeback.

You need to watch the Monument Records concert from 20 years earlier where he does all these songs as a younger man. His voice was even more thrilling back then. Especially great is "Mean Woman Blues".
