There is an editing error there. It shows Harry cocking the revolver into single action mode as he points it at the wounded robber, then it shows him pulling the trigger double action. Albert Popwell, the actor portraying "I got's to know" plays several different characters throughout the Dirty Harry series.
If you look at many of these scenes you will see there is no ammo in the cylinders. Some are hard to tell. others it is obvious depending on the lighting.

It was actually a .41 magnum- there was a shortage of .44s at the time. The frame is the same. The .357 frame is smaller. Almost impossible to see the difference, as a .44 is actually .429, whereas the .41 is correct.
