Why was it recommended to perform a streak method when isolating bacteria from infected leaves?

Why should we take 50%infected portion and 50% healthy portion of the leaf ? If we take 100% of the infected portion..what will be the outcome sir ?
Health looking part is already infected. Higher concentration of pathogen is in healthy part near the death edge. In dead part bacteria might already consume all nutrients and die itself, so border between dead and alive is best.
What kind of environment should one work under to isolate fungal pathogen?
Is it a sterile distilled water?
For the sub culture which media we used?
Same as for the early one, it depends what you are culturing commonly used media are, pda for fungal growth and nutrient agar for bacteria growth
