    This all depends on how you fill capsules. If you are adding the drug directly to the capsule manually, then you don't need to calculate the amount of filler. You just use excess filler on top of the drug in the capsule until it's full.
    Now, if you are making a homogeneous mixture of powder that contains filler and drug (preferably mixed with 3D or conical screw lab mixer), then you need to calculate the exact amount of filler and drug you require AND ensure all capsules are filled to a similar mass.
    After filling you do a mass spec to determine if you are getting enough drug in your capsules...you should get two peaks...one for the filler and one for the drug. Most automated capsule fillers incorporate a blender. Doing it by hand is when problems arise.

    what if I'm using a filler for the purpose of making the medication slow release? I think methyl cellulose is lighter per volume unit.

    Nice self explanatory video , my question what about if you have multiple excipients ( FILLER , GLIGANT ,LUBRICANT…..) how you do calculate the excipients quantity ?
