There are 3 ways. Talk to villagers with a large blue circle over their head, complete previous missions, but I think your problem is experience. Look at the locked mission, and you will see a number. Now look at the number with XP next to it in the top right corner of the screen. This if you haven't guessed, is the amount of experience you have. If this number is lower than the number in the mission's box, than the mission will remain locked.
