Depends on how any linebackers you are using. If you are using 3 then sure. 1 is contain, 1 is cursh and 1 is cutback. If you are using 2 then , 1 is contain and 1 is cutback and they try to vice the ball carrier. If you are using just 1 then he better be a star. lol
Question: I coach youth (11-12 yo). We run a 5-3 w stand up DEs who have contain responsibility. Would you still teach the the 3 LBs the same principles (contain, crush, cut back)?
Excellent question! And the answer is yes. Even if your stand up ends were LBs the fact still remains that once they’re on that line of scrimmage they are linemen. The reason those DEs have contain responsibility is because you want to funnel everything back inside. The last thing you need is a kick out on your end and a seal on your outside backer because once a back gets through that, he’s gone. Still teach that playside backer to get to that outside shoulder of that block and squeeze that run back to the mike. You also have to teach your backers how to properly scrape and not get picked up by linemen. All of this will really help you if any team is really good at running power
Who does the inside linebacker on the zone or man do they cover the running back or which receiver do they cover be cause there are no recievers that come up the middle
It depends on your defense. Your sam could be out in coverage on a TE or slot or the Sam could've been replaced by a Nickel back and is out in coverage. Your Sam could have shifted to the end of the line of scrimmage or blitzing off the edge. There are a number of cases where the Sam could be removed from the box. Same goes for the Will backer. In some defenses the Will may be removed from the box leaving the Sam and the Mike. Whatever the case may be, this is how two linebackers would handle a run play in between the tackles.

How would it change if I’m running a 5-4?
