- How to Set up HPLC calibration curve - Internal Standard Calibration Method is demonstrated in this video, we explain to you why and when internal standard are needed in the analyses. We also show you how to determine an unknown concentration of Caffeine in a tea sample with Paracetamol as the internal standard. The Shimadzu i-Series LC-2050C 3D was used in this demonstration, which has a photodiode array (PDA) as its detector. We also demonstrate how to accurately prepare a set of standards for internal calibration in order to obtain a good calibration curve.
This video is part 2 of the HPLC Analysis Tutorial series which provides a step-by-step demonstration of how to operate the Shimadzu LabSolutions software in order to obtain a calibration curve, starting from setting up a method file to run a realtime analysis all the way to processing the data in the postrun window.
