Hey Ed, how long and how hot have you found to be optimum for drying>
Most fruit should be cracker dry in 12 hours depending on your dehydrator and temp. I run mine at 55C/130F for a total of 24 hours, then store them in gasketed Sterilite/Tuperware with pre-dried silica gel. If they are too large to dry in that time, split them in half.

I prefer prints because they are easier to mail internationally without customs (First class international stamps). I find less gills in a print than swabs when receiving them.
I send swabs internationally all the time. No problems. I use business-size envelopes and orient the swabs vertically (I cut the long wood part halfway). Never had a problem with over 30 countries. You shouldn't see gill fragments on swabs. You should find a new vendor. That is someone who doesn't understand what they are doing and you will just as likely get a clone instead of spores.
@edwardgrand I did try that and I have a jerk postmaster. He's the only one that works there. If he feels a lump he won't send it. Returns to me with notes lol. Last one I just tore the cotton off and sent. That worked 😂
Can I sterilize 5ml cryo tubes with the lids on? I always do my media bottles with the lids tight and have no issues.

Hello Ed, thanks a lot for your wisdom and teaching! I'm into gourmet mushrooms and can adapt alot from your videos to my process. Your teaching about monokaryons is really exciting for me. This season I got a lot of spore prints from my mushroom community foreaged from the wild. Unfortunately they are pretty dirty as most of the people are not familiar with the process (too long on the aluminium foil, no burner prints, mushroom touching the foil etc.). Anyway I want to work with those prints and try to clean them up. Do you have any advice for this scenario? Water gellan plates, maybe even with some active charcoal in it? Serial dilusion? Most of my concerns are on the bacterial side, and I'm not sure if e.g. a serial dillution would help with that? I've seen your trap door tek video, but I wonder how to start i nthe first step with a dirty print.
H2O2: I been hearing people talk about spraying the prints with hydrogen peroxide. I've never done it, but maybe it works? AB: Gentamycin, Penicillium, Cephalosporin and and other antibiotics might help. Serial dilution: I've tried it, but unfortunately it dilutes the spores and bacteria together. Charcoal: Maybe, but I doubt it with spores. Trap door, cabinet sequestration and trenching: might help clean up bacteria after the culture is started. Race substrate/plates: you can see if the mycelium will outrun the contams on agar or substrate. Water/Gellan plates: Might help the spores germinate, but so will contaminating molds. Might help with the bacteria. There are a lot of options, but I would try to get better prints. Starting with dirty prints is a major waste of time. I would concentrate on the clean ones and maybe forget about the dirty ones unless you really need them.
@@edwardgrand Yes you're right, better to focus on clean prints :). But there is at least one dirty spore print from a really interesting phenotype and the mushrooms are not so often to find here (sparassis crispa), so I'm very happy to receive even dirtier prints and I will give it a try anyway to have as much genetics to work with as possible. I might try out the H2O2 spraying, serial dilution and water/gellan plates in a row to fight mold and bacteria simultaneously. I worked with a tek called puck drop (tiger drop 3.0) which seems also interesting for this because it is a brf substrate. Thanks for your advice!
I have reverted getting prints in some glass petri dishes that don't have a match. Got a box of 40 that were tops only for free, can't complain. Then just take another one and place on top and wrap. Also thinking about the parchment paper ironed to sterilize.
i love the prints but be careful where i store it! hahaha 😂 i love the look of prints and making art with it but i can use a print 25times(my current burma) from pre Covid era. swabs i get to use about maybe 5times for one? so i get more outta the prints but the work is different and storage of course. i also like to collect the prints cause they have character
My swabs come 10/pack, so I hope that will be enough to last me until I grow it again. Prints do have character, but I'm not sure they are worth the time and effort for me.
Big ups to you my bredda 🫡Sending you my greatest gratitude ! Concern: My monos/ spores sometimes never germinate. They’ll be no contam which is good but no growth. Sometimes I felt I was swabbing too damn light in attempts for solid monos. OR I thought my recipe needed altering for better germination even though I do get spores that germinate successfully as well. But my dumbass never thought I could just be swabbing too early in attempts to not swab too late haha I Needed this head slapper, I feel slow lol
There are many pitfalls. I have made all the mistakes I talk about, probably multiple times. That is why I talk about them to save others the same pain and suffering. :)
i’ve been taking prints with one or two desiccant packets in the plastic box. The prints come out fine, i’m not sure how necessary it is to have a super humid environment is for cubes.

Hi Ed, what kind of feedback are you getting from your customers regarding getting swabs through customs checks ? I have got spore prints through from the US, to Ireland, but never liquid culture, even my gourmet syringes from within the EU get opened and inspected, never ordered swabs.
I send swabs to many countries. Never had a problem. They fit flatly in a business-size envelope. They go straight through with the normal letter mail.

How many times can you use a swab ? Once you swab, moisture from the agar will be on the swab , this will no doubt detrimentally affect the spores. So, are swabs single use, or good for several plates but still only to be used on one occasion. Whereas a print can be scraped 20 times on twenty different occasions.
You should watch my other videos on swabbing. All your questions will be answered. You let the swabs dry before storage. You can use a swab multiple times by ripping off small pieces of the swab with tweezers. You can swab multiple plates with one swab. You can make >20 swabs from a medium-size cap. Swabs are cheap AF and store well in small ziploc bags.

I so wish you would do and sell prints on foil still because swaps are more problematic in sending via post internationally.Prints are best in my opinion.
It is actually better to send swabs. There is no metal to show up on the detectors. I send swabs all over the world. They fit almost flat in a business-size envelope. Never have a problem.

Missed it. Great stream Ed! Thanks for the early morning lessons! I only like spore prints because I can make swabs at a later date without having to regrow the fruit. Prints are a pain to make but I find them convenient for giving spores away. Of course you could just make more swabs from the fruit and not make the prints. Personal preference I guess. 🙂

As a novice I’m not sure if I’m getting spores on my swab from albinos or if it’s gill tissue
Swab as lightly as possible. Normally, spores will take 5-10 days to germinate and grow pretty slowly for about a week. If you see rhizo growth after 3-5 days, it is likely cloned gill tissue.
The only argument I can make for prints rather than swabs is potential genetic diversity. If I’m correct with prints I’d assume you’d inevitably get different phenotypes again assuming you’re using the whole print vs a swab where you’re probably only getting spores from a few gills. Other than that I’d pick swabs any day of the week for convenience, ease of use, and sterility. 9:03
That is definitely true, but I think the number of spores on a light swab would represent more genotypes/phenotypes than any one person could run in a lifetime. Most people end-up isolating a monoculture and narrowing it down to one possibility anyway. I love running multi--spore spawn these days, so that is a nice compromise for me. I'm not narrowing it down to one phenotype, but it makes the results in the tub more manageable.
@@edwardgrandthat’s exactly my thoughts. I could never see myself running the same exact print for more than a couple grows, because by the 3rd grow I’m already inclined to pull spores again. Swabs are so seamless, literally less than an hour from swab to plate. You can probably work the same print for a whole generation if you used tiny enough samples - I’m WAAAY to ADD for that😂
Love the show! I much prefer swabs but out of habit I always get prints from everything as well, it is a pain but I use a screen so nothing ever touches. I agree though that swabs store better and are easier to use for everything but wild mushrooms.
That screen trick is kind of cool. I've also used chop sticks balanced over the top of a glass to make MSS. I will add that to the list of cool things to do with spores. Thanks for commenting.

Michigan is not really north enough for a good view. I think I say them once in the U.P., but we were on a lot of mushrooms :) I think he might have been talking about the Sothern Borealis because I live in Thailand???

It just looks dry because it has the hull still on it. The grain is actually 'al dente'. Dryer is better when it comes to the surface. But you want the grain cooked properly. It's a difficult balance to strike.

I agree with you on how sad it is people buy other people's cultures and sell them. Thats the entire reason i wont sell gourmet cultures. I always just assume anything i create and give to anyone will be expanded and sold. Its disheartening, so i just stick to fresh mushrooms and give away my originals. So many people are low lifes these days always wanting to make money without doing any actual work.

Another Ed live. I’ll have to listen to the whole thing in a bit. My mon mon’s don’t do to well but the di-mons seem to do so much better.
Mon-mons seem to just make generic fruit in the F1. My di-mons often throw something pretty cool. Maybe the mitochondria in the monokaryon (recipient, 'female') greatly affect the expression? I noticed all my crosses with the Toque mono had Toque hats. Just a thought. I wonder if some of the cytoplasm and organelles get transferred with that nucleus from the dikaryon?

Completely unnecessary. Cracker dry, stored with silica gel in a sealed container is the easiest, most convenient and cheapest way.

On the topic of freezing: Brian at WTFungus overwinters his blocks, fruits them in the spring when they thaw. So it's at least very doable with blocks
