@@baloog8 She has been asked about prolactin increase and its relation to prostate cancer. This was her response: “The prolactin signaling cascade associated with prostate cancer is not an item in isolation. For it to promote cancer proliferation, synergistic effects with other molecules and pathways need to happen. Therefore, the assumption that an increase in prolactin completely negates an entire literature of studies and reviews on the benefits of melatonin in prostate cancer is flawed, IMHO. In addition, it is unknown what amount of melatonin was given even though the term high dose was used. Understand that there is a huge difference in effects that can be associated with the use of 30, 300, and 3000 mg melatonin.”

You Does the melatonin made by the pineal gland and released into the blood stream at night make it into the mitochondria of the cells of the body? Copilot Melatonin Explore Yes, melatonin produced by the pineal gland does make it into the mitochondria of cells. Melatonin is an amphiphilic molecule, which means it can easily cross cell membranes and enter different cell compartments, including mitochondria1. It’s synthesized in the pineal gland at night and released into the bloodstream, where it can then be taken up by cells throughout the body. In the mitochondria, melatonin acts as a potent antioxidant and free radical scavenger, which is part of its neuroprotective role1. Additionally, melatonin is also produced in the mitochondria of all other cells in a non-circadian manner, and the amount synthesized by the pineal gland is less than 5% of the total2. This endogenously produced melatonin influences glucose metabolism in cells2. You Since the mitochondria of the cell needs the stimulation of near infrared light to make melatonin is it possible that at night when the mitochondria can no longer make the melatonin to clean up oxidative waste from energy production that the pineal gland excretes into the blood stream the melatonin at night cause the mitochondria can no longer make it in the absence of near infrared radiation? Copilot The relationship between melatonin production and near-infrared light (NIR) is indeed fascinating. During the day, NIR from sunlight can penetrate deep into the body and stimulate the mitochondria to produce melatonin1. This process is thought to be mediated by the activation of specific enzymes and proteins within the mitochondria, such as cytochrome C oxidase2. At night, in the absence of NIR, the pineal gland becomes the primary source of melatonin. The secretion of melatonin by the pineal gland at night is part of the body’s natural response to darkness3. This increase in melatonin levels is associated with the body’s preparation for sleep and is a key factor in regulating the circadian rhythm4. The idea that the pineal gland compensates for the lack of NIR-induced melatonin production at night is an interesting hypothesis. While the exact mechanisms are complex and still being researched, it is clear that the pineal gland’s release of melatonin is crucial for maintaining the body’s daily rhythm and providing antioxidant protection during the night when mitochondrial production is reduced5. In summary, both sources of melatonin-mitochondrial and pineal-are important for the body’s overall well-being, with the pineal gland taking a more active role in melatonin secretion during the night. This dual mechanism ensures that melatonin is available to help neutralize oxidative stress throughout the 24-hour cycle.

Totally agree - I've been a virus skeptic since 2008. A germ (causing disease skeptic, even before that). and I have critically examined the science.....closer to $cience. Point being, Electron micographs do not prove conjectured particles causing disease. Growing a "viral' soup via cell culture is not true isolation. If we examine how any functional medicine or drug works on an alleged viral cell culture, the beneficial effect we are seeing is its effect on the antibiotic and anti-fungals added to the cell culture, alleged to contain viruses. So is how part of the self-fooling occurs. Indoctrinated, trained scientist assumes these standard lab protocols have been well vetted, however....only as far as the grant money flows. That being said, in terms of mitigating disease, either pro-oxidative therapies or upregulation of NRF2, or diet that does not perturb the glycerol-fatty acid cycle (mixing carbs and fats for example) will allow not to aggregate as much toxin beyond our bodies ability to deal with it.

8:43 she lost me when she indicated that mankind originated from some type of "primordial soup". Talk about religious dogma. It takes much more faith to believe that mankind crawled up out of a cesspool than to believe that mankind was created by a creator. These brilliant people must know that human life could not have evolved from chaos into this extremely complex system. from Claude AI "While Doris Loh presents a comprehensive overview of the relationship between melatonin, phase separation, and ATP production in mitochondria, it is essential to examine the existing scientific literature to validate these claims and explore additional perspectives. Phase separation and viral replication: Recent studies have indeed shown that various viruses, including SARS-CoV-2, exploit phase separation to create viral replication compartments or "factories" (Perdikari et al., 2020; Savastano et al., 2020). These membraneless organelles concentrate viral components and host factors to facilitate efficient viral replication and assembly. Melatonin and ATP production in mitochondria: Melatonin has been demonstrated to influence mitochondrial function and ATP production. Studies have shown that melatonin can protect mitochondria from oxidative stress, enhance electron transport chain activity, and increase ATP synthesis (Reiter et al., 2016; Tan et al., 2016). However, the specific mechanisms by which melatonin regulates ATP production in the context of viral infections require further investigation. Melatonin and reactive oxygen species (ROS) scavenging: Melatonin is a well-established antioxidant that can scavenge various ROS, including hydroxyl radicals and hydrogen peroxide (Reiter et al., 2016). By reducing oxidative stress in mitochondria, melatonin may help maintain optimal conditions for ATP production. However, the direct link between ROS scavenging, matrix viscosity, and ATP synthase efficiency, as proposed by Doris Loh, needs additional experimental validation. High-dose melatonin and viral infections: While some studies have explored the antiviral effects of melatonin (Reiter et al., 2020), the specific doses required to achieve these effects vary widely. Most studies have used melatonin doses in the range of a few milligrams to tens of milligrams per day in humans (Andersen et al., 2016). The extremely high doses (several grams per day) proposed by Doris Loh for regulating viral phase separation have not been extensively studied in clinical settings and may require further safety and efficacy evaluations. Melatonin and chronic conditions: Melatonin has been investigated for its potential therapeutic effects in various chronic conditions, such as cancer (Li et al., 2017), neurodegenerative diseases (Pandi-Perumal et al., 2013), and autoimmune disorders (Carrillo-Vico et al., 2013). However, the specific role of melatonin in regulating phase separation and cellular energy production in these conditions remains an active area of research. In conclusion, while Doris Loh's lecture provides an intriguing perspective on the potential of high-dose melatonin in regulating viral phase separation and promoting cellular energy production, further research is needed to validate these claims and determine the optimal dosing strategies for various conditions. As with any new treatment approach, it is essential to proceed with caution and rely on well-designed clinical trials to establish safety and efficacy. " and more in general about phase separation "Phase separation plays a crucial role in various cellular processes and organelle formation within human cells, including heart cells (cardiomyocytes), liver cells (hepatocytes), and neurons. Here's an explanation of how phase separation works in these cell types and its relationship with organelles, mitochondria, and phospholipid bilayers. Membraneless organelles: Phase separation is the driving force behind the formation of membraneless organelles in human cells. These structures lack a phospholipid bilayer and are formed by the condensation of specific proteins and nucleic acids. Examples include: a. Stress granules: Found in all cell types, these structures form in response to cellular stress and contain RNA-binding proteins and stalled translation initiation complexes. They help protect RNA and regulate translation during stress conditions. b. P-bodies: Present in all cell types, P-bodies are involved in mRNA degradation and contain RNA decay enzymes and RNA-binding proteins. c. Cajal bodies: Found in the nucleus of most cell types, including cardiomyocytes, hepatocytes, and neurons, Cajal bodies are involved in the assembly and modification of small nuclear ribonucleoproteins (snRNPs) required for splicing. Mitochondria: Mitochondria are double-membrane organelles that do not directly involve phase separation in their formation. However, phase separation can occur within the mitochondrial matrix, where it helps organize metabolic enzymes and regulate their activities. For example, the enzyme acetyl-CoA carboxylase (ACC) can undergo phase separation in the mitochondrial matrix to form punctate structures. This phase separation is thought to regulate ACC activity and, consequently, fatty acid synthesis and oxidation in cardiomyocytes, hepatocytes, and other cell types. Phospholipid bilayers: Phospholipid bilayers are the main components of cellular membranes, including the plasma membrane and the membranes of organelles like the endoplasmic reticulum, Golgi apparatus, and mitochondria. While phase separation does not directly form these membranes, it can influence their organization and function. For instance, phospholipid bilayers can exhibit phase separation between liquid-ordered (Lo) and liquid-disordered (Ld) domains, depending on the lipid composition and temperature. This phase separation can affect membrane fluidity, permeability, and the distribution of membrane proteins, which can have implications for cell signaling and transport processes in cardiomyocytes, hepatocytes, and neurons. Neuron-specific phase separation: In neurons, phase separation is involved in the formation of specific structures and the regulation of synaptic functions. a. Postsynaptic densities (PSDs): PSDs are protein-rich structures at the postsynaptic membrane of excitatory synapses. Phase separation of specific proteins, such as PSD-95 and SynGAP, is crucial for the formation and organization of PSDs, which are essential for synaptic transmission and plasticity. b. RNA granules: Neurons contain various types of RNA granules, such as transport granules and synaptic RNA granules, which are formed by phase separation of RNA-binding proteins and mRNAs. These granules are involved in the transport and local translation of mRNAs at synapses, which is essential for synaptic plasticity and memory formation. In conclusion, phase separation is a fundamental process that contributes to the organization and function of various cellular structures and organelles in human cells, including cardiomyocytes, hepatocytes, and neurons. While phase separation is not directly responsible for the formation of phospholipid bilayers or mitochondria, it can influence their organization and function, as well as the formation of membraneless organelles and neuron-specific structures."
