      We usually do not instruct clients/people on specific methods. However, it is our recommendations based on products similar to the ones in question are: for bread and baked goods - AOAC official method 922.06 and for ice cream and frozen desserts - AOAC official method 952.06.

    Hi there. I have questions regarding my test using Soxtec 1043.
    Is it necessary for the solvent to boil or it is enough when the refluxing solvent drop into the cup?
    What if solvent in some cups, let's say 3 cups out of 6, boil and the rest don't? What could be the reason for this phenomenon?

      Hi, Setya. The method requires visible evidence of active refluxing in every condenser tube by observing the collection of solvent in the side arm of each tube. There are several reasons that may contribute to the cups not boiling including the cups, chips, plate, etc. Please refer to the manufacturer instructions. I hope this helps!

    Hi,
    Can you suggest me low sample weight requirement analytical methods. I need to find protein, carb and fat content ; sample weight is 6-8 g only.

    I need to test %fat in fish bones and scales. Is it possible to use this method?? please advice which AOAC standard that i need to follow. Thank you.

      Kanageswary Sockalingam The video presentation is an overview of the method, and we cite references that pertain to our applications. When sample matrix are different than the ones specified, we urge the client to check the literature for the specific applications.

    If I want to test the %fat on animal carcass, is there any official method I can refer to?

      The official method for determination of crude fat in meat is the one we cite on the video - AOAC Official Method 985.15 for fat in meat.
      Before the fat analysis, it is very important to prepare the sample from the carcasses. See literature for the sample preparation.

      Flour could be analyzed by Soxhlet as well as by acid hydrolysis methods depending on the sample. It is the individual responsibility to read the methods and decide the best one for the sample type to be analyzed.
      For flour samples please refer to:
      Soxhlet method: AOAC Official Method 2003.06
      Acid hydrolysis method: AOAC Official Method 922.06.
      The percent fat method we describe on the videos is an overview or summary of determination of %fat by Soxtec based on the official methods we make reference during the presentation. Before attempt any of the techniques, it is mandatory to read the entire method that applies to the specific sample matrix to be analyzed.

      As mentioned on the referred video, the determination of total lipid of a sample by organic solvent extraction is specific for certain matrixes. For the total lipids determination in milk, the official method is:18th ed AOAC Official Method 989.05 - Fat in Milk, Ether extraction method. JAOAC 71, 898(1988).
