HPLC Troubleshooting - Column Back Pressure

  • In this video I talk about column back pressure in a liquid chromatograph. The HPLC will generate very high back pressure, much higher pressure that we will encounter in everyday life. The pressure is caused by the pump forcing the mobile phase the element through the column and so the high back pressure occurs between the pump and top of the column. The high pressure is not something we set it just happens and its a function of the particle size in the column, the diameter of the tubing, the length of the column, the viscosity of the mobile phase and the temperature at which we are running the system at.
    The back pressure is measured in different units depending on different HPLC systems, sometimes it will be in psi, usually American systems use this, it could be in Bars which most Europeans system use.You need to be careful with setting your pressure because of setting the maximum and minimum cut outs.
    What would be a normal pressure will be something you have to determine for your system, there is no accurate or right answer but its important that you set an appropriate upper limit that will stop the system if the pressure goes too high. Then actual pressure then could give us an incorrect answer if we find that something has gone wrong.....

    Lol what even is back pressure. I was hoping for someone to at least explain what it is.
