You can plan your journey something like below, Hyderabad to Kolkata by train Kolkata to guwahati by train Guwahati to shillong you have to go by road only, you get many cabs....I think they charge 250 per person Covid test is mandatory before entering meghalaya, so I suggest you take RTPCR(it is valid for 72 hours) test in guwahati only because if you take your test in shillong then you need to stay quarantine in shillong until you get the report(which will take ~2 days) and unfortunately if it is positive then you have to be quarantined for 14 days and the stay/food is very expensive in shillong....There is no covid test required in assam, west bengal.
You can also take a direct train from Hyderabad to Guwahati. It's a weekly train, I am not sure of the frequency post Covid rules. From Guwahati there are cabs as well as regular buses to Shillong.

In northeast, we don't call it ghat roads. This is only applicable to South India. Western and eastern ghats
