Yes they've spoken in multiple interviews about how they've read the books, they're reading Messiah now to prepare for the next one, when they say "I wanna know what happens" they're talking about the next movie, not what happens in the books
From a lot of their interviews it seems most of them did read Dune (I don't think Denis would've hired them otherwise) but not the other books after that (especially the 2 after). And I'm not sure if it's because the questions from interviews aren't that deep or if the interviews don't really understand thematically what Frank Herbert intended with the Dune universe, but it doesn't seem like most of the actors understand and can analyze the main themes of Dune, particularly the environmental influence of human civilization, the evolution of the human mind over technological advancement, the consequences of religious worship, etc. As an avid reader of speculative fiction and sci-fi I get so excited when iconic scifi works like Dune get adapted onto the screen, but then it's disappointing when you see that the actors and/or creators can't get into the literary analysis that these works demand. It's as if people don't even consider sci-fi a literary genre beyond spaceships and fancy technology.
