    The most incredible thing about Karl Barry Sharpless winning the Nobel this week isn't the work he won it for, but the fact that when he won a previous Nobel 21 years ago he spoke about what his next Nobel prize would be about.

    KBS was just starting up as a prof at MIT when I was a grad student. He had 6 grad students at the time and was investigating organo selenium chemistry.

    Thanks for the nice presentation of click chemistry and Bioorthogonal chemistry. The following are the doubts.
    1. Click chemistry involving copper to link azide and alkenes is fine in non biological system
    2. Whereas in physiological system glycan get bound to azide. Later reactive strained alkenes may react any molecule, why alone to glycan coupled azide in a living system.
    3. Does azide bind to only glycan. Does it not bind to protein or nucleic acid or lipids.
    4. Is it similar to coupling a fluorescent dye or an enzyme to an antibody in ELIISA . Or real time PCR.

      From my knowledge, I can clarify the point 3 that they've done a lot of experiments and found that every time the azide only bind to glycan due to some biological specificity based on Lock & key theory.

    Sufex click reaction is another type of click reaction given by Sharpless team.
    And they say this reaction is a sleeping beauty. Much more useful in the near future

    hmmm i don't understand how combining two molecules together is different from lets say, monomers turning into polymers? is it that useful because you can stick azide and alkyne to *other* molecules that don't easily combine to get those attached?

      The dye tags the cell. If there's any spot that's colourless, that's the infected cell. Depending upon properties of dye or bacteria the other way around is also possible.

      In the case of molecules having functional groups with oxygen, Cu2+ oxidises the groups and destroys the molecules and in bio systems, protiens are the molecules that are used in such reactions, their native structure and function gets destroyed.

    Click chemistry is not only about copper catalyst.....other metal catalyst can also be used in chemistry.....this statement is clearly wrong that click chemistry cannot be used in biological system... I myself doing my PhD in click chemistry
