It’s Butcher who despises Homelander, but despite having several opportunities to kill him, Homelander has never taken that opportunity with Butcher. In a weird and severely screwed up way, he’s the closest thing to a friend Homelander has, because he’s brutally honest with him and doesn’t hide his contempt for him, which he really respects. Butcher is one of the few people who’s ever been ‘real’ around Homelander.

I always loved that moment where Hughie stood his ground and looked more furious than scared when Homelander made sexual remarks about Annie. I don't think Homelander had any respect for Hughie until that moment. He still despises him, but I don't think he viewed him as someone with no backbone after that. He knows Hughie has zero chances against him, yet is impressed with his courage.
Nearly everything about Homelander being taken off guard or whatever could probably be explained by this, He's self absorbed and in the belief that he's basically God. So much so that he probably doesn't put effort into most things, Underestimates nearly everyone because ''he's the perfect being'' and just generally doesn't seem to be trained in anything really, Because he thinks he doesn't need anything but his powers.

Only nuke can kill homelander from human weapon arsenal or some kind of bio weapon.
