The chest was closed when he found it and only opened when luffy was next to it, they just never showed it to us. So no, shanks never knew about luffy being nika or joyboy reincarnated. (I found out about this some time ago so I don’t remember all clearly but if this helps)

Luffy died because of kaido, but the gear 5 gave luffy all his energy back. Luffy used his powers to drain kaido's energy, which allowed him to quickly defeat kaido.
