They're hunting for "matsutake" mushrooms that are the most expensive kind in Japan and really look like those depicted in this anime :)
In Japanese, a matsutake mushroom actually has a metaphorical meaning of penis due to mutual resemblance in shape, and I believe it's obvious that the author deliberately indicated the metaphor by "hunting for a matsutake unicorn".
