Berlin ZOO is in the Mitte on the outscirts of Tiergarten Park. There is Zoologischer Garten Bahnhof just outside of the Zoo so you can go where ever you like with U and S-Bahn in minutes. I think Potsdamer Platz is 5-6 minutes ride, Brandenburg Gate is 10, Alexander Platz is 15...If you wanna walk you will need like at least 45 minutes to get to the central landmarks.
