Hi. What fixative do you recommend for making parasitology slides out of stool samples?
Working in a medical lab. I’m trying to teach some friends how to recognize some amoeba…

    What about:
) UV cure clear epoxy resin
) Vacuum pot + Alcohol, Acetone, or Formalin
) 3D printed cover glass standoff frames
    ) Balsam or synthetic balsam

Hi I really enjoy the videos, very informative, I wonder if you have some advice to seal urine samples to make permanent slides. Thanks

I tried clear nail polish but when it turned hard the clear nail polish turns into yellowish and not clear anymore,so when I look at the slide with clear nail polish mount under the microscope light can't be able to pass that's why it's so dim when I look at it.

What mounting media do you recommend for preparing permanent textile fiber slides?

I'm literally just starting out in microscopy having purchased a Tomlin DM620 Pro digital microscope, and your videos will be my go to source for educating myself. Am I right in thinking that the subject matter for mounting with pva should be as dry as possible to begin with, and can it contain a small amount of moisture? Thanks

PVA glue is water-based, so it does not matter if there is some moisture present in the specimen. Other mounting media (resin-based ones) should be dry or at least be in a solvent that is compatible to the mounting medium.

I would imagine the PVA glue would work nicely

20% water? This is probably why I didn’t had good results. I didn’t had enough water.

I'm still worrying about quality of speciment after mounting.
I have speciments of stomata (gaurd cells of plant leaves).
Does the cheap mounting media can affect the speciment quality?
    Thank you!

Not price or quality counts, but the type and how well the specimen was prepared. Different mounting media and preparations for different specimens. For plant cells, use PVA mounting medium (or other water based), otherwise the organic solvent will remove the green color (chlorophyll) of the plant cells.
