No, calculating the rate of transpiration using a potometer isn't a required practical. If an investigation involving the rate of transpiration comes up, you will be given enough information to be able to work out the answers - they are likely to be mathematical skills, i.e. calculating rate (distance/time). I hope that helps :)
It definitely is the first time you see it, as can involve a bit of Maths in questions. Just think of it as a way to measure how much water the plant is losing through the leaves - if you can visualise it as the plant pulling up the water then that might help!
It depends if the stomata are open, but normally yes, the plant is losing water continuously. The stomata will close if it is very hot, dry or windy i.e. in conditions where the plant is losing more water than it can replace by osmosis in the roots. The stomata may also close at nighttime, as no photosynthesis can occur in the dark.
