  • This is very simple method for calculation of Optical Density of your IHC images using ImageJ Software.

    I don’t know if you still answer questions on here, but I’m having an issue once I finish calibration. Upon measuring an image, the max value fir OD shoots super high (285521.668, to be exact). I think its because the MGV fir that data is equal to 0, and the function goes exponential. Is there a way to fix this so that it doesn’t deviate so greatly?

    The method was very simple and great. I am using it to measure protein expression in DAB stained immunohistochemical slides. But, can you please provide any reference of a published article that used this method? It would be really helpful. Thanks.

    Can you tell please, how to know the optical density for mast cells (stained with toluidine blue) in order to classify them in different types according to their staining intensity

      You just need to convert your image into grey scale...so that your stain will be dark black which you can measure

    Hello! Thanks for this very helpful video! But I have one question. I obviously got different mean gray values in my picture than yours, so how can i find the known OD values to calibrate my image??

    Hello. Thank you for this very helpful tutorial. I have a few questions: Why every time I open ImageJ and do the calibration of step tablet image I get different mean gray values? Why even though I take the same mean gray values as previous measures with the corresponding O.D I get every time different numbers in the calibration curve formula?

      may due to minor difference in the selection of area......i haven't faced that kind of problem...

    what if my IHC picture has both H and DAB brown? what should I do to calculate OD? is there a easy plug in?

      no there is no any plug in....but u have to select the region

    Hello, do you know if I need do this kind of calibration to measure the grayscale of muscles? Or just the measure scale is enough?

    hello thanks for this video, i have a question. I want to quantify IHC in the aorta can I quantify the intensity of staining too by this method? I am comparing the protein expressions but I don't know how to do measurements.
