There is no secret on the quantity of memory being used. It is just MSX's very clever slot architecture + rom mappers at play. It is very common to find MSXs running with 2 or 4Mbytes of RAM. In fact, you could go up to around 40Mb, following strictly the standard. This is throughly documented all around the internet and books.
On the other hand, a Gradius reverse engineering would be something else. I'd vote for the reverse engineering, but, if possible, not only aiming at the Easter Eggs, but also on overall game structure and tricks and magics from Konami.
