In oily solutions of direct inoculation method why we use fluid thioglycolate medium for detection of aerobic?? It is mainly for anaerobic

Thioglycate medium basically an enrichment medium and it contains sodium thioglycate. Sodoum thioglycate reacts with the oxygen present in the medium, and reduce oxygen level to create anaerobic environment. In this condition aerobic bacterias can grow only on the top or the upper side, where aerobic bacteria get oxygen. But inside the medium or at the bottom only anaerobic bacteria can grow because there is no oxygen and strict anaerobic environment is there. So we can check the presence of aerobic and anaerobic bacteria by using the same culture media.

Explain Sterility testing for IV fluids.

Sterility testing of IV fluids are done by membrane filteration method. IV fluids are first filter through membrane filter and then the membrane is placed on suitable culture medium. This is the very basic and common method used for IV fluids.
Other then this IV fluids are also tested for presence of pyrogens through in-vivo pyrogen testings.

Autoclaving is the method used for sterilization of Parenterals, but if the formulation contains and thermolabile substances like enzyme, hormone or any other proteins, then sterilization is achieved by aseptic filteration through membrane filters.
