Basically he’s laughing at corrupted Stewie with the “Giggity giggity” because he got rid of corruption by doing something that relates to him as a character, which is the “Giggity Giggity”. I think it’s just that none of the other characters try the stuff they like.

I just realized, since Lois didnt look like your average darkness victim, they were obviously mutated into a monster-like form, bringing it down to Stewie during Rotten Family, it is then shown IN twinkle where stewie has a monster-like form with a ton of tentacles, whenever quagmire uncorrupted stewie by storing the darkness in his arm, later on in airborne, he got stabbed, making it so that the darkness he stored was free from his arm, now stewie's past corrupted form got taken down to Quagmire, as seen in Airborne he is ALSO a mutated monster. Could this be a coincidence?
After he stabbed by Stewie his gun and his arm is gone because the glitch sliced or ripped his arm off and btw the extra the reason why in when Stewie was getting freed was because when quagmire had his arm cut there was a some glitch on his lost arm so instead he traded half of Stewie glitch in to his body and that’s how Stewie got freed
