Principle and Theory - It is based on in vivo experiment. Here we have to involve complete experimental animal for the experimental purpose. When there will be a sensation of pain at the base of the tail of mice it will try to flick the tail and that will be noted as a response of pain. The same process will be carried out with control and experimental animal group.
Pain- pain is an unpleasant feeling, which makes us uncomfortable and reduces the physical as well as mental alertness. Although pain are also useful for us because it acts as a warning signal and it warns about something uncommon inside or outside our body. If the pain is minor it may be tolerated but if the pain becomes severe it has to be managed at earliest.
Analgesic- Analgesics are the drug which is used to get relief of pain. Although there is no such drug which completely removes or reduce the pain by its root. The analgesic drug only suppress the sensation of pain or increase the threshold of it. The reason for pain perception is the genesis of cyclooxygenase (COX). COX is of 02 main type- I and II. The main target for NSAID (non-steroidal anti-inflammatory drugs) is to selectively inhibit the COX-II
    Procedure-
    (1) Weight the animal and divide them into 02 groups and marked them as a (1) Control and (2) Experimental group
    (2) ake the basal reaction time by applying the current in the form of heat to the wire attached with the screw. So that the tail of the rat will receive the stimulus for pain. That should be around 02-03 cm of the tail length
    (3) When the heat is applied heat will produce and pain will be felt by an animal, and then it will flick its tail, which indicates that animal is feeling pain.
    (4) This time (basal reaction time) is recorded. Normally rat withdraws its tail within 3 to 5 sec. A cut off period of 10 to 12 sec. are observed to prevent the damage to the tail of the experimental animal.
    (5) Take 03 to 05 basal reaction time for each animal at a gap of 05 minutes to confirm the normal behaviour
    (6) Inject normal saline to control group and given a drug to the experimental group and note down the reaction time at each 15, 30,60, 90 and 120 minutes. (Depending upon the object you have received)
    (7) Calculate the percentage increase in reaction time (Index of analgesia) at each time interval and note down.
