Hello madam, this is good video about TEM technique. I have been working with TEM from last 15 years and I would like to point one thing here that is ultramicrotomy( sectioning) is done after the sample preparation ( sample embedded in resin) then staining with heavy metal and finally viewing in TEM. If you are talking about cryoultramicrotomy then sample preparation is not needed. Otherwise the normal sequence is always first sample preparation- ultramicrotomy-staining-viewing.
