Hello there do some npc mission in any city in the map if you complete all the NPC quest in the city you will unlock a Gwen Pool Mission in that particular city :)

In short you need to collect gold bricks by completing npvs mission to unlock gwenpool missions.

Its an unlocked mission. Do quests around Manhattan and eventually you will get it
