A lead is a clue that police use to solve crimes, a Lead is also what is attached to a collar around a dog's neck to stop them from running away. "Police said there were no leads" therefore they ran away because there were no leads (to stop them running away). You can interpret it slightly differently but the core of the joke remains the same.

I've watched this show for over a decade and I can honestly say, without a doubt, Ryan Stiles' best bit.
That’s your opinion, but I think Wayne and Colin are just as good as he is!

Ryan does it better in the acts he is awkward at like hoe downs, sideways scenes or stuff like reversing the scene
It's hard to point out a singular instance where he was his best. At least for me. Hard to pinpoint something like that over a career that has lasted decades. But maybe it stands out to you more because you love the movie as well. Either way, his bit had me clutching my stomach. Best part is, it wasn't overtly sexual in order to get an awkward laugh. Reminds me of the old days.
