He decided it was time to entrust it to the next generation or something and then told his body to shut off. Even Hashirama's cells were affraid of Hashirama so they did and he died.
On a more serious note it most probably was the same desise that the Sage of Six Paths and Naruto got. It's caused by having insane amounts of chakra in a human body (the sage was half human half otsutsuki but with a chakra reserve that's high even by Otsutsuki standards). Shit probably killed him. Wow. Didn’t know all of that. He’s one of my favorites because his personality and because he just seemed kind of cool, but that adds more to the epic nature of his character. Also, Minato is one of my favorite characters as well.
Hashirama's ONLY weakness was his personality. He wanted the best for everyone so much that he underestimated the cunning and ruthlessness of other people. This is why he was unstoppable with Tobirama by his side. He needed a trusted confidant who could prepare and make back up plans just in case someone tries to betray his older brother. Alone they had "blind spots" but together they completed one another. I so wish we had a movie that delved more into the Naruto-verse politics so we could flesh out the relationships between Madara and Izuna, and Hashirama and Tobirama. I would have found that so interesting. We did not get enough and it's important for fans to understand how people felt when the leaf was founded. And we definitely needed an explanation as to why people distrusted the Uchiha even after a century of Konoha's founding. I refuse to believe it's only because they were powerful. The Sarutobi, Akimichi, Nara, Yamanaka, Hyuga, and Hatake clans all distrusted the Uchiha years after Konoha's founding AND after they excommunicated Madara, and I wan to know why. What was behind that mistrust and what was fueling it? The movies, novels and fillers gave us some insight (e.g. they continously placed clan before village, unlike the others. Which means that most were incorrectly prideful and selfish) but none of this is 100% canon. I hate speculating on this stuff without more evidence.
I dont think Hashirama underestimated anybody's cunning or ruthlessness. He was so increadibly op and in a league of his own that he didnt care what the other person was like. Cause they were never going to be able to beat him anyways. He fought the Uchihas for years and never lost once. And the Uchihas were the most cunning and ruthless lot.

Do you remember when Hashirama wanted to give away the tailed beasts for free? Tobirama prevented this and Konoha charged the other countries for each tailed beast, except for the Sand Village who already captured Shukkaku. All that extra money was extremely important because it allowed Konoha to rebuild after the First Great Shinobi war faster than the other nations, which saved their economy and allowed them to maintain their strength above all the other villages. Additionally, Hashirama wanted Madara to simply accept the new way of things. However, Tobirama doubted Madara's dedication to peace. He knew that Madara was too much of a warlord to let his past go so easily. Under Madara's rule, he would want to force the other villages to submit to Konoha's dominance. And after just a few years Madara left the village and soon after attacked. Possibly contributing to Hashirama's early death. To be an effective Hokage one has to be more than just a strong Shinobi. They have to be a diplomat, an administrator, an economist, they have to balance ALL of the affairs of the village including the ugly messes which arise, they have to have a sense of foresight to prepare the village from possible trouble in the future, and one has to be a strong Shinobi. Hashirama couldn't do all of these jobs alone, hence why he relied on his younger brother Tobirama. Hashirama may have been a great diplomat but he wasn't good at reading people's intentions. It's always better to stop a war from ever happening than to prepare for one. This is what I was referring to, and this is what Hashirama continously underestimated. Kishi, the creator of Naruto, probably modeled Hashirama after those people most of us may know with really big hearts who are always trying to please people. They are so kind with only benevolent intentions that somtimes they remain naive to those who seek to exploit and hurt them. That's why if these people are lucky enough, they'll have a sibling, spouse or friend who acts like their protector. Constantly looking out for danger and are suspicious of others. Making sure that other people are not a threat. This is the role that Tobirama served for his brother Hashirama. All of this is also elaborated on in the novels. This turned that Uchiha into a powerful shinobi but also turned him into a hateful, bitter and single minded person. And since the Sharingan was also the entire base of the power of an Uchiha, any powerful Uchiha was naturally mistrusted by everyone

Shit like this is the reason why I feel like Naruto should have had some extraordinary about him not involving Kurama. Naruto essentially had nothing special from being an Ashura reincarnate unlike Hashirama.
