The old woman remembered, as a little girl, seeing a young geisha, then decades later (as now a middle-aged woman) seeing an exact doppleganger of that same young geisha (because both times it was Daki). The madam put two and two together and realized the old woman's story was about Daki and since she doesn't age she must not be human.
Right… but just one correction: they are not geisha. Geisha are female entertainers (although some did sell their bodies). These girls are prostitutes or courtesans who also have other entertainment skills.

Like Director said she's basically the HBIC, sometimes with more power than the owners of the House that they serve as Geisha. Both get sold or offered free of charge to the houses they serve like slaves back when. Sometimes boys as well as girls since dudes were into that. Like the feudal lord Nobunaga Oda was either gay and still had kids for his lineage or he was Bi and had one or two male consorts). The mistress who was killed, it's their job to manage the house and the girls in it, while usually their husbands (if they have them) own the house and work the money and security.
