on the 3-3 stack the teams we have played brought the third backer as an overhang and he is blowing up a lot of plays to side the running back is on the will also rotate the safety down to cover the y receiver
I don't run 3rd level RPO's, but if you are playing against aggressive safeties that trigger on the mesh point, then I would run the RPO off the Safety to the single WR side with a 5 step post.

Would your read key be determined by the alignment of your RB?
If you mean alignment as in either left or right of QB, our read key is always on the same side as the mesh point with the QB+RB
