It does make sense in a way because even though Rosé is the main vocal, Jisoo is the lead vocal and doesn't have a dance position. I'm also pretty sure that I've heard artists say that it's easier to sing with a hand mic or that it sounds better so it would make sense for the vocal who doesn't have a dance position to have the hand mic. I have also seen performances in which Jennie and Lisa are the ones with a hand mic and I figured it was because they're the rappers so I can't be sure what the actual reason is.

It's probably because she's the lead vocal and doesn't have a dance position. Rosé is the main vocal but she's also lead dancer, Jennie isn't technically a lead or main vocal and objectively a better dancer than Jisoo (Jisoo is a really good dancer but they're all amazing dancers), Lisa is obviously main dancer so it wouldn't make sense to give her the only hand mic.
