- Explaining the process of plant tissue culture processing for columnar type cactus ( Echinopsis pachanoi) and buttons
Processing step before surface sterilization
I did Echinopsis hybrids from seed and got callus from it which turned into LOTS of plants all on its own.
Awesome video! Can’t wait to see how they progress
Do the successful tissue cultures have a similar growth rate to new seedlings?

Is this a home lab you're working at? Seems like a fancy setup!
Home lab videos coming out this month;)
cool
Hey I got a question if you don't mind answering. The PH of my media starts dropping over time, especially once I stick a plant into the vessel.
For example, pre autoclave of 6.10 becomes 5.8 post autoclave. After cooling, I stick a plant in and let it incubate around 26 with regulate photoperiod 16/8. I check the media's PH after a week and it drops to like 4. I still get growth from my plant, but I was thinking if the PH stayed consistent the plant would grow even better.
Do you find the same with your plants, and if so, do you personally try to buffer the PH somehow? Or not care and let the plant do its thing.
Thanks!
Not care and let the plant do it’s thing. Most literature does not adjust so I actually assume the pH change is due to the celular signaling and metabolites. I find no need in any of my work
@@cactifanatici thanks for confirming!
@@cactifanatici Appreciate it! :) Thank youu
how do you make the media? what are the ingredients? greetings from brazil.
MS based media either autoclave or microwave tek
@@cactifanatici how long is it microwaved for? Do you add anything else to the MS
@@cactifanatici you should do a video about Ms based media tutorial
