It depends on the card. Some of the cards automatically trigger. Others require an action by the player, and the player making the action has to meet the trigger requirement.
It is pretty clear (as long as you understand the difference between the 2 and 3+ player game symbols, which I made a mistake on and noted in the video). Once the trigger is flipped, the players shown on the effect portion get the benefit.
