I just got into DAW's(Digital Audio Workstation) and plugins and audio interfaces for my cheap ass guitar to sound better, and I believe that's what people use to isolate vocals or any other instrument for that matter
For most songs people use the tracks from Rockband/guitar hero games cause they can isolate every instrument there. Some songs have official instrumental versions available too. However for the case of this song in particular its not in gh or Rockband yet so I'm not sure
