According to concept art he was based on the Nickel Plate Road S2 Berkshire, which is the same thing as the as 1225 in real life. Of course he was made into a 4-8-4
Side note, just to clarify, cuz I've seen at least one person in these comments confused, Samson is a 4-8-4, aka the Northern Type (or Niagra if you prefer the New York Central term, or Heavy Mountain if you prefer the Santa Fe terminology)

pm 1225 is a n-1 berkshire and s-2 is nkp 765 so samson is the prototype of the nkp s-2
