    If we had an error to our serial dilution which there's a lot more growth happened in the latter dilution 10^-4 to 10^-6, how can we troubleshoot this kind of error, or what could be the reason for this? sorry for this kind of question-

      There could be errors in labelling or pipetting or could be cross-contamination.

    Thank you so much for this explanation... please I further need help, I want to apply bacteria population ranging from 10^6 to 10^8.. I did the serial dilution and plating, after counting the petri dish with 10^-7 gave me a cfu of 30 and because I used 0.1ml for the plating, I ended up having 30,000000. Please how do I get a bacteria population range of 10^6 to 10^8. Thank you

      The population that you got which is 30,000000 falls between 10^6 and 10^8.

    I got everything, but I have one question. When we take 1ml from each tube and transfer it to the next, the concentration of all of them decreases to one 1ml, but the last tube will have 1 ml more bcz it wont be tranfered to the next tube. so how do we bring its volume equal to the rest of the tubes?

      Concentration does not depend on volume. Example: Prepare a 1000 mL solution of NaCl with 1mg/mL concentration. If you transfer 500 mL to another container, its concentration will still be 1mg/mL only.

    If I were to use vial 3 and 4, how will I standardised / average them? From the reading, they are 2,600,000 CFU to 3,500,000 CFU respectively. Will the average be 3,050,000 CFU?

      Choose the one with countable CFU.

      @@MicrobiologyMantra thank you. Yes, both have countable CFUs, as per your illustration. Though the computed ranges are wide, that's why I am seeking guidance.

      You cannot take average of two different dilutions. You must take the average of two plates from the same dilution.

      I don’t have any particular reference for this topic. I made this out of my own practice knowledge and resources from the Internet as well. There is lot of information available on this topic on the Internet. I suggest you to summarise and make notes. That will make you more familiar.
