Not sure if you need a trap on that , if there is a back flow preventer on the pipe then you will be good but if not you will have fumes coming up that new drain pipe
With a tank that big I would suggest a pond pump for water changes. You can buy adapters for them with garden hose thread. Then you can buy an adaptor for you sink to thread on the same hose and run the water straight to the tank, never mess with a bucket or suck on a hose again!

We do have a one way valve on that pipe. They actually have the whole basement cut off from the main somehow after they had a flood in the past

I was thinking about that, but honestly if we just got an inline bulb hand pump. it would be all we need now
