If electric force is attractive and external agent is moving the test charge away from the source charge. Or when force is repulsive but external agent is moving the test charge towards the source charge. In simple when work is done against the electric force then there will be increase in potential energy. If there is a movement in charges due to own attractive or repulsive force then potential energy will be negative.

Electrostatic force is negative gradient of electric potential. Easy meaning is - force is in the opposite direction of maximum change (increment) of potential w.r.t. space. And the magnitude of the force is the rate of change in potential in that direction.
