In my opinion, the system quest is more literal. Not like it would kill him if he doesnt kill them, but THEY mean to kill him and if he doesnt kill them, he'll die. And as he is its host, it wants him alive, so it gives him rewards as an incentive to stay that way.
The subtitles are kind of weird sometimes, I think as a localization thing. Anaki and nii-chan are basically elder brother/respectful and brother, but nobody really says "hey, brother", so "hey, jing-woo" seems more natural. Aniki translated as boss is a bit weird, but is close enough in meaning to get the point across, I think. The other comment explained what the potion did, but in case you forgot it was a reward for defeating the snake, he quickly dismissed it at the time because of the debuff effect of -30% or whatever strength. He did just drink poison, but it was for the buff not for healing. He has a general 20% damage reduction now. Willpower is a passive damage reduction skill that activates at low health.

I've enjoyed watching your reaction videos. Some of what you're saying about jinwoo in the beginning is cultural differences. You have to remember this is written by a south Korean author. Their country has been under constant threat of war for the entirety of the authors life. They're raised with a mentality pf enjoy the peace while it lasts but be prepared to fight for your life at any moment. At least thats how life was described by some friends i had who were from there. And this anime seemded to kind of emobdy that sentiment. Be well
