In fact if we go back more than a millennia, it's widely speculated that the land Shanghai is on was still mostly underwater. It was only due to the sediment deposited by the Yangtze River that this land exists at all. The trade routes going in the direction of Shanghai were probably heading for Suzhou or Nanjing - both cities actually existed back then and are pretty close to modern Shanghai (100km&300km respectively). They also both used to be capitals during various dynasties so there were pretty good reasons to go there as well.

A quick google search showed Shanghai was already a major trading city by the 1700s, which predates the Opium Wars by over a century.
