    Studies presented as science are really not science at all. They just pose a question and whether it merits further investigation as to the actual underlying biologic function. Don't base your living choices on 'conclusions' from studies unless you want to be a statistic.

      By reducing Reactive Oxygen Species (ROS) primarily producing in complex 1 and 3 in ETC.

      I think its because it upregulates AMPK. It's also known to negate the benefits of exercise so I would consider it a net loss. This is all part of the David Sinclair hype train in which everyone lapped of his garbage like mothers milk and he got rich. The next air to the throne is Bryan Johnson. More sales with no robust outcomes.

    I went low carb lifestyle. Taking metformin (Dr prescribed) I had hypoglycemic attacks twice a week. I stopped the stuff, still have 3 full bottles.

      Studies have shown it inhibits anabolism(muscle building) by about 10%. That’s the reason Peter Attia quit taking it at one point. I don’t know if he’s still off of it or not. Unless you’re doing body building, when you’re younger 10% probably won’t matter much. But as you get older, especially past 65, it becomes more important because it gets so much harder to build muscle.

      I took it for awhile however decided to get off of it /p(my choice for both). I didn’t find it made much difference. Also, I’ve heard that it can deplete B12 so I did make sure to supplement B12 and/or B-Complex.

    Metformin lessens the risk of glucose inflammation but does not address or treat the main cause which is addiction to highly processed foods and thus the dietary overconsumption of glucose/fructose the consequences of being chronic tissue damage associated with aging. There are other factors into aging, but a highly processed food diet is a major cause of aging. The natural and more direct way to lessen the damage caused by high glucose blood levels is a more active lifestyle, better sleep habits, PLUS a low carb/starch, lo-Omega6 fat, hi-Omega3 fat, hi fiber diet which in turn would reduce aging and extend the lifespan without medications or expensive surgery. However, most medical doctors are not trained in nutritional/dietary science or in physical therapy or how to handle addiction with highly processed foods.

    You said it’s not necessarily the researchers’ fault, but the simple fact of him getting so excited with the media attention, as well as all this talk of repurposing based on a study that goes against randomized control trials, shows a handful of redflags…

    Berberine is supposed to be as good as metformin for those with type 2 diabetes. It would be nice to see the exact same studies done on it for anti-aging.

    Compared with metformin, berberine exhibited an identical effect in the regulation of glucose metabolism, such as HbA1c, FBG, PBG, fasting insulin and postprandial insulin. In the regulation of lipid metabolism, berberine activity is better than metformin.

      Yes, I switched from metformin (and it was free to me) to berberine and it has helped me lower my a1c and has helped me lose weight.

      Maybe, but Berberine (and many other phytosupplements) is by far more expensive than metformin.
      From my local pharmacy I can get 30 tablets (1000mg/tbs) metformin with less than 3 euro, this is 60 days supplementation!

      @@hristosstrihas4010 You have to do what you have to do, meanwhile, Liposomal Berberine is available, minus the side effects of Metformin for the most part and will blow it away on both counts, A1C and Lipids.

    Working out hard doing cardio and strength training 6 days a week makes me feel good and healthy. That is the only supplement I trust. No other snake oil for me.

    I took metformin to experience if it works after 2 weeks of taking metformin I quit because I wasn't feeling good, I had low energy, I was depressed and confused. short after I learn that metformin destroys the good microbiome and inhibits the absorption of vitamin b12.

    Metformin ruined my dad kidneys and did not control his type 2 diabetes as well as Berberine did, which as a bonus reduced his cholesterol and improved liver markers too and its prescription free

      Most people wouldn’t have to be on these drugs if they just lost enough body fat and built some muscle. So sad and unnecessary.

      Some people who have diabetes have higher risk for kidney issues so not sure how you’d know it was the drug or his diabetes, etc., that ‘ruined’ his kidneys, etc.

      @@BlahBlahPoop617 Pretty much hit the nail on the head. Uncontrolled glucose levels (honestly, anything ever over 140) are like little knives to the periphery capillaries. This damage accumulates and is the reason we lose toes, vision, suffer ED, and do organ damage. That said... metformin is not great for kidneys, so if you already have kidney issues, you need to come off metformin (nurse)
      Also, I'm 8-10% body fat. Very well muscled, but if I am not insanely strict with my carbs and exercise timing, my glucose stays in the 200s. Shitty genetics are still a thing

    There was an Interventions Testing Program (ITP) study that evaluated metformin's effects on mouse lifespan and aging. The results showed that metformin alone did not significantly extend lifespan in these mice.

    There are a bunch of subsequent studies that show metformin reduces long covid, improves the microbiom, delays aging through lengthening telemeres, improves cardiovascular disease, and attenuates varios hallmarks of aging. I was pre-diabetic 5 years ago. I changed my diet, added moderate excersise and began metformin. Today , my symptoms of high bloodsugar have all disappeared, and my fasting glucose is always in the mid 80s. I still test every morning. Metformin is cheap as dirt, and safe. Even if you put aside the longevity benifits; just the fact that thru diet, excersise and metformin, my bloodsugar is optimal and i am now at my college weight. 57 years old with my college body back!, makes metformin very well worth it for me.

    @ 06:26 "we believe". Ironic that a researcher should pick this verb phrase since it is what almost all of us also fall under in making our choices. Ask anyone what scientific proof they have and you're likely to get ramblings that amount to nothing more than hearsay. It's a sin we're all guilty of.

      One problem is that people don’t necessarily carry a bibliography or list of their references in their heads even if they have read the studies. If you ask me what my evidence for X is, I’d have to get back to you on that after looking everything up (again), which is too much trouble for most folks.

    Doesn't berberine have similar benefit with fewer side effects than metformin?
    That's what I take. Has helped me lose 5 lbs. and reduces carb/sugar cravings too.

    Watched an KOL using it for anti aging but he shrivelled like a prune with lots of wrinkles in a couple of years. Frankly he is approaching 60 something but he was a good looking middle aged man. Now he looks fragile and wrinkled

    Will done, I was not so luck, always when doctors prescribe one drug you need another to protect from a side of first, am sure that how drug companies made drugs (one drug and by the way you will need other one as will ) also PPI's drug to protect your stomach, from drugs that can damage the lining of your like same blood thinning drugs, also stops you stomach for absorption of B12 🙏🙏🙏🤔

    I know this turns off many people (pandemic fatigue). Interesting study: Metformin reducing chances of long covid. That can be interesting considering many are experiencing multiple unmitigated reinfections with Covid these days

    I’m not clear on how metformin is expected to “push back death by age-related diseases” other than type 2 diabetes? Do they already understand its mechanistic contribution to longer life-span in that very specific population?

    I can see why the interesting Metformin. Nic the other study you mention on longevity used several drugs to reduce a fatty thymus I think and it used Metformin. When 80% of the risk factors for cardiovascular dies ease and stroke are metabolic including diabetes.It makes sense that if you reduce those risks you will live longer. Metabolic disease and diabetes is sky rocketing most likely due to the food we eat and lack of exercise. This mostly a lifestyle exacerbated by genetics problem., with environmental concerns added.

    One group of researchers has been looking at it for yrs as part of a cure or prevention for aging and or life extension. Watched a good doc video on it a few yrs ago and read some articles.

    I have seen patients taking dihydroberberine, 100mg after dropping 500mg of metformin, and their results where better. Also, reports shows metformin could cause peripheral neuropathy which from there causes diabetic foot ulcers

      Metformin definitely worked not well for my dad's diabetes and kidneys . Berberine worked much better and didn't affect his kidneys. Most doctors know Metformin affects on kidneys hence why l kidney disease people avoid it

      It will still be. It’s common domain, so unless the companies change the molecular structure, it will still be very cheap. Even if they do, the current version will still be very cheap.

    Metformin very noticeably but reversibly reduced my libido. Anyone know the mechanism? (I am metabolically very healthy, HOMA-IR around 0.5.)

      It reduces testosterone. It had devastating effects on my ability to walk and I am female over 60. Imagine what it would do to men's testosterone? I have zero confidence in our so-called medical system and even more skeptical about research professors.
      Nick we look to you to restore our faith that there exist honourable scientists who are not easily bought to be shills for big pharma.

      I would say, MET down regulates MTOR via upregulation of AMPK.
      This move the body into a catabolic state. Such state would naturally be found in a long-term fasting food-deprived situation. I observed the same during a weight loss period. Sex and proliferation is not the priority if there is no food.
      But why on earth would you take MET with a HOMO-IR of 0.5?

    I was on met for for quite a few years. It causes neuropathy. This is one of the big problems with it. I had the flu like symptoms from it. My doctor told me to push through it and yes, after while I was on met Forman and 70-30. What a shit show made you hungry all the time gained so much weight got so unhealthy then they put me on blood pressure medicine. The fatigue was huge. I walked out last year started getting better. It’s over a year later now no more strokes June was the last month Doing so much better with nothing I didn’t mention the statin, but that gave me swollen legs two weeks after quitting the statin my right leg had gotten pretty normal not so swollen. The other one took about nine months. I don’t believe what Doctor say anymore that’s why I listen to you, trying to find the magic pill that will make me better at some point I won’t need anything yet. Berberine was a big thing for me. It helped so much now I take probably a dozen supplements and doing better getting my energy back slowly very slowly, my doctor keeps wanting to put me on a blood pressure pill, the funny part is they don’t even know what causes your blood pressure to be high yet they wanna put you on. I read about everything would not take a blood pressure pill there’s natural ways to control it yet. It’s still high. The nitric oxide is huge from what I read I eat beats every dayyet still have high blood pressure keep looking for things for that one have the best day ever and keep up the great work Nicolas I watch everything you do you help me very much

      I had a stroke as well and I've tried every supplement known to man, yet can't get off blood pressure pills. I'm fit, lift 4 times per week, walk to gym and grocery store and it still rises when I get off the pills.

      It seems like you can avoid neuropathy by supplementing B12. Diabetes also causes neuropathy.

    Perhaps the longevity crew should take a look at the research on an even cheaper drug that can't be patented: lithium and its alleged anti dementia affects. Been a while since I've heard anything about that one

      And also other mental health benefits. Apparently in regions where the water has more lithium, there are fewer mental health disorders. Even microdoses could help in the medium term, so you could track down the highest concentration water you can regurlarly obtain and drink that daily. If anyone from Portugal is curious, found a study of portuguese waters that analised lithum concentrations and Vidago and Pedras Salgadas had the highest. Vidago has less sodium, so thats what I buy. Just 250ml per day is comparable to a microdose of lithium for some mental health disorders.

    I've spoken to many centennials. I live in the Caribbean isles of Trinidad and Tobago. Some smoked for decades, they drank soft drinks, ate fry foods, barely took supplements. And they all lived in lightly forested areas. And the secret to their long lives was so simple. All of them were not obese also. People of African/Indian/Mixed decent.
    They ate once or twce a day. Small portions. Rice, meats, vegetables. Did some kind of work around the home/yard. Rested during the day in intervals. Made sure to rest early, no meals prior for an hour least. Ate fruits also. Normal small amounts. No candies. Chocolates yeah. Drank tea, bush tea, coffee. All of them also was exposed to sunlight doing little gardening or stuff in the yard/land. When it's extremely hot, they all kept inside. So mostly early morning and evening exposure, when it's cooler, as it's the tropical region.
    So my conclusion, the key to long life is what you consume, how much, giving the body time to repair/recover, no strain on the digestive system. One thing, they all ate around 6pm. went to bed 7 to 9. No supplementation, if any, an occasional vitamin if they got from a family member. So through foods, they had to be getting essential nutrients.
    I guess your digestive system, not putting to much stress on it, and little exercise.
    The elders that lived to their 60s to 80s, they all either smoked heavily, were diabetic, and were not eating as the centennial fully. One had heart problems from birth.
    Many individuals on drugs, I've known or do know, eventually developed new syptoms or conditions down the years, sickness never healed and rid of, but masked.
    From my own experiences when I was in my 20s to mid 30s, I was muscular, slim, BMI on spot, bodyfat was within 15 to 20 percent. I never used to get sick, even amongst sickly people. Can be in rain and full sunlight, in the bush amongst dirt, creatures. Used to eat twice to three times a day. Small portions. Breakfast was very light. No oily foods, margerine, soft drinks, alcohol, smoking. Vegetables, fruits, meats, rice. Always active, lifting weights, doing martial arts. AND no stress from woman, till recently. Stress is another factor towards aging. Sometimes it ain't woth the expence of your longevity and happiness.
    I've slacked off alot. Getting back on track. Can't cheat or skip your progress. It will be harder to go bak on the rails from derailment. Hopefully this comment, can help people. I'm no expert. Just sharing some things off my experiences and self analytics thereof.

      The life expectancy of someone in Trinidad and Tobago is 74.87 years, which puts it at 96th. This is far below even the US, which is pretty bad for an OECD country. Just saying.

    Metformin has a recall in 2022 for containing NDMA a carconogenic ...not good for the stomach also ...the toll on the digestive
    system can be quite bad for some people ...dont think i will be taking any ...i,m on Berberine now and its ok .

      Agreed Metformin wrecks my stomach even after a month of 250mg ( a low dose) stopped taking it.
      I'm also very sensitive to Wegovy as well insane acid reflux.
