Initial unwinding is carried out by helicase which separates two strands of DNA at Ori site. However, Topoisomerases cause DNA strands to unwind after replication starts ahead of replication fork....so technically both helicase and topoisimerase are correct

Congrats to you for your selection. I have no idea about the facilities and faculty of department of Baotany at central university. I did my pg from university of kashmir and it was really a good decision. I easily qualified all Botany based competitive exams (NET, JRF, SET, 10+2 lecturer exam and was also topper of Assistant professor written exam) after completing my PG.. Whereever you do your PG, you goal must be to become expert in the subject you are studying...and success will definitely embrace you.. Anyways, Best of luck for your life....
