You can clear the Barrow as soon as you escape Helgen. Almost everything you fight in the game levels with you so each challenge is at your difficulty. Over time, certain enemies will be level capped. So bandits will be one hit kills later in the game, but the dragons continuously get more powerful. Basically the game is designed so any quest you take, should present some challenge. The game is designed so challenge is picked by the game difficulty level, as opposed to certain areas being higher level content.
