You load loading control in every well?
Loading control is not loaded. They are proteins, mostly cytoskeleton proteins, that are expected to be present in all the samples in 'equal' amounts. This means that if you are seeing differences in the intensities in this protein for the different wells/samples, then you did not load your samples with equal concentration.

At minute 3:33 you say that there is no GTP bound to the tubulin. This is not the case because alpha tubulin is still bound to GTP.
That doesn't matter because the GTP bound to alpha is unusable and is basically restricted inside alpha tubulin..
