Thanks for your quesiton! Changes in media color often come from exposure to air, but media color changes can also come from contamination. This happens with bacterial contamination, which causes the pH to decline and begin changing the media to an orange or yellow color. The media often also has an increase in turbidity with bacterial and fungal contamination. Some fungal contaminations can cause the pH to increase and become more pink rather than the neutral pH red color, but this is less common. Please let us know if you have any other questions!

That's correct! The sample enters through the dull side facing up, and the filtrate exits through the glossy side facing down, as shown in the video.

If your sample has high particulates, yes, you can use Millex HPF: ms.spr.ly/6057YAZ0t Otherwise, if your sample is not heavy on particulates, Millex PTFE with HDPE housing should work (even with glacial acetic acid): ms.spr.ly/6059YAZ0v. What is the error when the blue solution does no settle down like in the video instead it mixes with the fuel ?

The blue capture solution attaches itself in the fuel water phase, where the microorganism is located. If the blue solution is not properly shaken, it will not be able to capture the microorganism in the sample. Additionally, the blue solution will settle to the bottom due to the previously mentioned fuel water phase, unless a glass bottle is used, which we don't recommend. There are many variables that can contribute to how long cells are viable for in -80˚ C storage, including cell type and the type of cryopreservation medium that the cells are stored in. Generally, cells will not maintain as high a level of viability when stored in -80˚ C vs standard storage in liquid nitrogen. Great question! We don't have a specific protocol for LAL testing of syringe filters. However, there are kits available, such as this - ms.spr.ly/6059cjeht - that can be used. (Please select the correct country and language in the top righthand corner of your screen.) Filter endotoxin-free water through the syringe filter, then test the filtrate to see if it contains endotoxin. The product information sheet has instructions on how to use the kit. Note that this kit is for research-use only. There are other LAL/endotoxin kits as well, a quick online search will give you some options. If your lab needs to comply with a compendial method (e.g. USP, EP), make sure that the kits you are using are compliant with the method described in the monograph.
