Ion Channels as Drug Targets - Medicinal Chemistry 1.18
The ion channel protein family is involved in almost all aspects of life. And because of how important the ion channels are in human physiology, they are related to many diseases and being able to control and regulate the channels become really important. Currently, the ion channel family represents one of the largest target classes for existing drugs.
--About me--
I am a computational chemist in the pharmaceutical industry. I created the Mole Man Chem channel to explain some basic medicinal and biochemistry concepts because I believe the best way to relearn a concept is through teaching.

G protein-coupled receptors (GPCRs) as Drug Targets - Medicinal Chemistry 1.17
Просмотров 4,9 тыс.Год назад
Drugs that target G-protein coupled receptors or GPCR account for near 30% of the global market share of therapeutic drugs. The high interest and large market share are because of how important GPCRs are in many diseases. Right now, there are over 2000 different mutations in 55 GPCR genes that have been characterized and linked to over 60 human diseases. Based on how those mutations can change ...
Receptors as Drug Targets (An Intro) - Medicinal Chemistry 1.16
Просмотров 6 тыс.2 года назад
Receptors are proteins, that by far, the most important drug targets in medicine. Because of the receptor's role in signal transduction, they are very valuable drug targets for treatment such as pain, depression, and even Parkinson’s disease and many other problems. If we can regulate the receptors, we can potentially regulate the downstream domino effect, or the downstream biological effects t...
Orthosteric Drugs And Allosteric Drugs - Medicinal Chemistry 1.15
Просмотров 3,7 тыс.2 года назад
There are two types of drugs on the market: orthosteric drugs and allosteric drugs. Orthosteric drugs bind at the active site of the target and directly compete with the endogeneous substrate. One example is the ATP competitive inhibitors that directly compete with the binding of ATP. Allosteric drugs bind elsewhere other than the active site or the orthosteric site, and they don’t directly com...
Suicide Substrates/Mechanism-based Inhibitors - Medicinal Chemistry 1.14
Просмотров 10 тыс.2 года назад
Suicide inhibitors or mechanism-based inhibitors are a type of highly-specific irreversible inhibitors that targets and hijacks the normal enzymatic reaction mechanism to inactivate the enzymes. They are relatively unreactive in cells or solution until they bind to the enzyme active site. For most of the irreversible inhibitors, the reactive functional groups are already present when the molecu...

Transition State Analogues/Inhibitors - Medicinal Chemistry 1.13
Просмотров 8 тыс.2 года назад
A chemical reaction will go through a high-energy intermediate state called the transition state. One strategy that drug discovery scientists came up with is designing an inhibitor that resembles the structure of the transition state. Also, this inhibitor could potentially bind strongly to the active site, even much stronger than the substrate or the products. This type of inhibitors is called ...
Enzyme Inhibitors (Reversibility and Inhibition Mode) - Medicinal Chemistry 1.12
Просмотров 4,3 тыс.2 года назад
Based on the reversibility, enzyme inhibitors can be categorized into irreversible and reversible inhibitors. Irreversible inhibitors would bind to the enzymes by forming much stronger interactions and “permanently” modify the enzyme. Reversible inhibitor binds to the enzymes by forming relatively weaker interactions or reversible covalent bond. When the inhibition is reversed, often times the ...
How did Pfizer/BioNTech win the COVID-19 vaccine race?
Просмотров 1,1 тыс.2 года назад
As of the beginning of 2022, only three Covid-19 vaccines are authorized or approved for use in the United States. They are from Pfizer/BioNTech, Moderna and Janssen Pharmaceuticals of Johnson&Johnson. As of December 28th, 2021, according to WHO, there are more than 300 COVID-19 vaccines in pre-clinical and clinical development. So, how did those three pull ahead and become the leaders in the r...
Enzymes as Drug Targets (An Intro) - Medicinal Chemistry 1.11
Просмотров 9 тыс.2 года назад
Enzymes are essential for life, and dysregulated or mutated enzymes could lead to diseases. Enzyme-targeting drugs make up about 47% of marketed small-molecule drugs. Besides how important the catalytic activity of enzyme is, another reason that they are frequently targeted for therapeutics is that they are relatively easy to target or they are more druggable compared to other biomolecules. Mos...
Conformational Selection Model vs. Induced Fit Model - Medicinal Chemistry 1.10
Просмотров 3,5 тыс.2 года назад
Both induced fit model and conformational selection model describe a flexible ligand-binding pocket. But the key difference between these two models is when the conformational changes happen. In induced fit model, the pocket conformational change happens after the ligand binding. In conformation selection model, the conformational change happens before the binding and the ligand selects which c...
What is the controversy over Aducanumab/Aduhelm for Alzheimer's?
Просмотров 1,8 тыс.2 года назад
In June 2021, the FDA approved Aducanumab/Aduhelm as the first new drug for Alzheimer’s disease in 18 years. It sparked controversies, surprises and even disappointment among scientific communities. Upon approval, three FDA advisers resigned due to the lack of evidence that the drug is effective. One month later, FDA Acting Commissioner Dr. Janet Woodcock requested an investigation to look into...
Proteins as Drug Targets: Microtubule Inhibitors - Medicinal Chemistry 1.9
Просмотров 3,6 тыс.2 года назад
Microtubule is an attractive target for anticancer drugs because of its important role in mitosis. In fact, it has been one of the most successful chemotherapeutic targets. Mitotic or microtubule inhibitors would disrupt microtubules and inhibit mitosis of cancer cells because cancer cells are able to grow and eventually spread through the body through non-stop mitosis. Cancer cells tend to div...

Thank you for posting this! Something I'm a little confused about: you stated around 1:04 that a peptide bond is formed via "a dehydrogenation or condensation reaction". Isn't dehydrogenation the removal of JUST hydrogen? So, it wouldn't actually be a dehydrogenation reaction in the case of a peptide bond forming?

id say both are likely and are not mutually exclusive, in fact id say that it is absurd to assume that selection can occur without the degree of variation (of one at least) still being capable of performing the necessary catalyses. Id also say that over time selection would move towards a lock and key level of fit. But ive been wrong before :) 10/10

This was really helpful :) I'm a new PhD student starting a project about targeting GPCR-mediated extracellular acid sensing by tumor cells and I'm just starting to learn why GPCRs are such common targets of drugs. In the future, maybe the exact GPCRs responsible for binding protons can be identified and targeting them can limit tumor cells from positively responding to the low acidity conditions they generate!

I didn't care about the after taste, I just needed relief from the pain and fever.
