Well I mean if you think about it GRU and the one doctor made a lot of experiments what if they weren't just using those on minions choosing certain once to enhance certain abilities in their body when undergoing stressful situations which would explain why GRU nor the doctor have shown signs of weakening even after aging. It would also explain why the doctor sometimes goes into coughing fits maybe he took something that damaged him a little in the process.
