GFP Production - Part 9B: Calculating Protein Molecular Weight from an SDS-PAGE Gel
Просмотров 1 тыс.4 года назад
This lab is part of a set of experiments to produce green fluorescent protein (GFP) in E. coli. For this video, we will be using Excel to calculate the normalized protein migration distance and native and denatured GFP molecular weights based on known protein standards. Refer to Part 9A for instructions on how to extract data from a gel (ruclips.net/video/IYcmYoajbgM/видео.html). This experimen...
GFP Production - Part 9A: Measuring Protein Migration Distance in ImageJ

This lab is part of a set of experiments to produce green fluorescent protein (GFP) in E. coli. For this video, we will be using ImageJ (imagej.nih.gov/ij/) to obtain data on protein migration distance. This will ultimately be used to determine protein molecular weight for our GFP native and denatured products (see the Part 9B). This experiment is part of my Bioprocess Engineering Lab (CM4125) ...
GFP Production - Part 6A: Cell Removal by Centrifugation

This lab is part of a set of experiments to produce green fluorescent protein (GFP) in E. coli. For this lab experiment, we are separating the E. coli cells from the liquid media prior to further purification of GFP from the media using chromatography. This protocol was developed in-house, but uses materials from EDVO-kit #303: Exploring Biotechnology with Green Fluorescent Protein from Edvotek...
GFP Production - Part 5: GFP Cell Culture

This lab is part of a set of experiments to produce green fluorescent protein (GFP) in E. coli. For this lab experiment, we are starting a cell culture to produce GFP using the E. coli that we had previously transformed. Following this experiment we will purify and analyze the GFP that was produced. This protocol was developed in-house, but uses materials from EDVO-kit #303: Exploring Biotechno...
GFP Production - Part 4: DNA Gel Staining
Просмотров 2814 года назад
This lab is part of a set of experiments to produce green fluorescent protein (GFP) in E. coli. For this lab experiment, we are staining our DNA gels containing previously generated colony PCR products, and evaluating the results. Following this experiment we will have confirmed whether the GFP gene was successfully inserted. This protocol is based off of the protocol and kit from Edvotek, EDVO...
GFP Production - Part 3: DNA Gel Electrophoresis

This lab is part of a set of experiments to produce green fluorescent protein (GFP) in E. coli. For this lab experiment, we are running DNA gel electrophoresis on our previously generated colony PCR products. In the next lesson, the gels will be stained to visually confirm that the GFP gene was present in our transformed E. coli. This protocol is based off of the protocol and kit from Edvotek, ...
GFP Production - Part 2C: Agarose Gel Prep for Electrophoresis
Просмотров 8804 года назад
For this experiment, we are preparing agarose gels for electrophoresis. Following colony PCR to amplify our GFP gene, gel electrophoresis will be used to visually confirm that the GFP gene was present in our E. coli. This experiment is one section of a single laboratory session that also includes visual analysis of microbial transformation results and prepping samples for colony PCR. This proto...
GFP Production - Part 2A: Transformation Results
Просмотров 1,3 тыс.4 года назад
In the previous experiment (Part I), we transformed E. coli to express green fluorescent protein, GFP. For this follow-up experiment, we will visually determine whether the plasmid was successfully inserted into E. coli using UV light and will calculate transformation efficiency. This experiment is one section of a single laboratory session that also includes prepping samples for colony PCR and...
GFP Production - Part 1: Microbial Transformation
Просмотров 1,8 тыс.4 года назад
For this experiment, we will be transforming chemically competent E. coli cells using pGFP, a plasmid that contains genes for ampicillin resistance and green fluorescent protein (GFP). Transformants will be selected for the presence of the plasmid using LB-ampicillin plates. In addition, transformed bacteria will be examined for GFP expression with and without IPTG to demonstrate the utility of...

    "There's a space here so it will do our averages automatically" What do you mean by that?

    • @rebeccaong2820
      @rebeccaong2820 Год назад

      The cell referencing shifts when you copy in Excel. So, if, for example, you copied cell C8 that contains the equation =AVERAGE(C3:C4), and pasted this into C9, the equation would shift down one row to read =AVERAGE(C4:C5). Since we copy two rows down it changes to AVERAGE(C5:C6), which matches where the replicate data is located. Extra info: If you want to lock cell referencing, you can use the $ symbol. By typing =AVERAGE($C$4:$C$5), this would keep the equation referencing cells C4 to C5 regardless of where you copied it. You can also lock just the column ($C4) or just the row (C$4), depending on your needs.

      @@rebeccaong2820 Good to know! At first for time 0 I tried to do my 3 replicates and then copy all the code across, however, it didn't give me averages (I didn't have a gap, I just had all averages stacked together hence why I asked)

    Can Visio be used to create complex P&ID with parameters? (i.e. pipeline and equipment properties)

    • @cinnamonfern
      @cinnamonfern 3 года назад

      Yes - you can use shape data (4:00) to specify parameters for pipelines and equipment. There are some built in fields depending on the shape, but you can also make custom fields. There are instructions for how to do this here: support.microsoft.com/en-us/office/add-data-to-visio-shapes-09272394-5243-4e1b-bcfa-425a8b4d1ce2

    Can I draw and solve material and energy balance problems including the calculations within Microsoft Visio?? I was searching for this for a long time and concluded that it has something to do with shape data but I still can't figure it out completely yet 😔😔 I hope you can help me!

    • @rebeccaong2820
      @rebeccaong2820 4 года назад

      You can draw the flowchart for a material and energy balance problem, and you can assign masses and enthalpies to streams, however I would not recommend using Visio to run the calculations (I don't even know if this is possible). Visio is a flowcharting software, not a process simulator so it is not really designed to run calculations. I would recommend either exporting a stream table with the data from Visio into excel (assuming you added this to the streams as material properties), or just manually input the data into Excel, skipping Visio entirely. Then run your calculations in Excel.

    • @shehabsoliman
      @shehabsoliman 4 года назад

      @@rebeccaong2820 Thank you so much for your response, that's what I managed to do and by using some flowcharting software add-in to excel it has become an easier process. However, I was just hoping to exploit Visio's diagramming capabilities in chemical engineering applications & problems instead of referring to the ordinary way to do this via Excel or Aspen Hysys or Plus, and so on. But thank you again for your answer. Appreciated!
