Ardent Scientific provides chromatography eLearning videos in High Performance Liquid Chromatography (HPLC). Each video is part of a training course and consists of animations with a voice over describing the content. While the videos can be viewed in any order, it is recommended to view them in the order as they appear in the playlist and preferably view them in order of their numbering i.e. Fundamentals of HPLC - videos 1 - 36. The videos will cover all aspects of HPLC. It is best to view the videos in 720p resolution on tablets.
Dr Gerard Sharp is the author of the course content and has more than 25 years experience in gas and liquid chromatography. In the last 12 years Gerard has developed course content and presented short HPLC theory and practical classroom courses across Australia and New Zealand. Gerard has a B.Sc (Hons) in synthetic organic chemistry from Monash University and a PhD in Analytical Chemistry from the University of New South Wales, Australia.

Why is important to know retention factor?
Because the retention factor shows the true retention of a compound i.e. the degree of interaction with the stationary phase. The retention time is only an indication.For example, if you connect a large length of tubing between the column and the detector, the retention time increases but the actual retention has not changed. Also the retention factor is useful for problem solving because it is a ratio of the time the component interacts with stationary phase over the time the component spends in the mobile phase. If the flow changes, the retention time will change but the retention factor will remain the same. If selectivity changes (for example the temperature or mobile phase composition or the column), the retention factor will change.

The more percentage of the organic modifier is in the mobile phase (more hydrophobic), the less chance the components interact with the stationary phase (hydrophobic).
